Metabolic stability studies of in vitro
Among the causes of failure in the in vivo use of a bioactive molecule there is the metabolic inactivation. The stability of a molecule can be easily estimated through a simple “in vitro” assay by using purified hepatic fractions in which the enzymes responsible for degradation are present.
The assay is performed by using S9 microsomial fractions or hepatocytes from different species. The percentage of unmodified parent compound is determined through HPLC analysis. The characterization of metabolites is performed by mass spectroscopy and/or NMR analyses.
